COURSE: ONE AND TWO DIMENSIONAL DATA PROCESSING AND ANALYSIS IN TOPSPIN

CREDITS: 01 

INSTRUCTOR: Clemens Anklin

RECOMMENDED PREREQUISITE: Laptop with Topspin 4.3 installed preferred (contact Clemens.anklin@bruker.com for assistance with installation or for help with license)

OBJECTIVE: To be able to open, process and analyze (not interpret) one and two dimensional NMR data obtained from a Bruker or other spectrometer. 

PROGRAM: 

This course will introduce the students to opening, processing and analysis of one and two dimensional data sets. All the steps are presented by the instructor and will be repeated by the students on their computers. We will use example datasets that are part of TopSpin. Alternatively students can bring their own datasets. 

Topics: 

Day1, Session 1 (8:30 – 10:00)

  1. Overview of Topspin software. This includes a presentation of the main user interface elements such as menus, buttons and pop-up function (0.5 hours). 
  2. Datasets will be opened from within Topspin, from the data browser or from ZIP files (0.5 hours). 
  3. Introduction to apodization, fourier transform and phase correction. (0.5 hours)

Day 1, Session 2 and 3 (10:30 – 12:30 and 14:00 – 16:00)

  1. Step by step process of apodization, fourier transformation, phase and baseline correction for 1D and 2D data. For 2D data sets the concepts of zero filling, linear prediction and NUS (non uniform sampling) reconstruction will also be introduced. ( ca 4 hours) 

Day 1, Session 4 (16:30 – 18:00)

  1. Peak picking, integration and deconvolution will be covered in this data analysis section (1.5 hours). 

Day 2, Session 1 (8:30 – 10:00)

  1. Continuation of peak picking, integration and deconvolution will be covered in this data analysis section (1.5 hours). 

Day 2, Session 2 and 3 (10:30 – 12:30 and 14:00 – 16:00)

  1. Advanced processing and analysis topics: Multiplet analysis, spectrum simulation (DAISY) and experiment simulation (NMRSim) Relaxation experiments and other topics. (4 hours).

Day 2, Session 4 (16:30 – 18:00)

  1. Q&A and practical exercises (1.5 h)
